Undo whatever you did to incurr the penalty and ask for a reinclusion.

First things I'd do is make sure it's a penalty. Lots of times you hear about penalties when it isn't the case and either there's just been a drop in rankings or someone has messed up.

Has your site been deindexed by Google - do a site operator check site:yoursite.com
Check the robots file, the no index meta elements and make sure no one has left anything in there that shouldn't be.
Are there any messages in Google webmaster tools.
